      壳寡糖(Oligochitosan,OCS)是氨基葡萄糖和N-乙酰氨基葡萄糖通过β-1-4糖苷键链接的低聚物,其表现出了诸如抗炎、抗氧化、促进生长等生物活性。目前壳寡糖已经广泛应用在人类或者禽类健康养殖上,但是有关其在水产中的应用还十分缺乏。本文通过在基础试验饲料中添加0(对照组)、200 mg/kg、400 mg/kg、800 mg/kg不同剂量的壳寡糖投喂珍珠龙胆石斑鱼(Epinephelus fuscoguttatus♀× Epinephelus lanceolatu♂)四周,综合分析壳寡糖对石斑鱼免疫调控作用。本研究首先检测了壳寡糖对鱼体内免疫及抗菌相关酶活的影响,结果显示投喂壳寡糖后,能显著提高珍珠龙胆石斑鱼谷胱甘肽过氧化物酶、溶菌酶、过氧化氢酶、碱性磷酸酶、谷草转氨酶和谷丙转氨酶酶活力。其次,壳寡糖的添加能显著提高了珍珠龙胆石斑鱼头肾占比。为进一步探究壳寡糖对珍珠龙胆石斑鱼的免疫调控作用,本研究通过实时荧光定量PCR检测珍珠龙胆石斑鱼头肾的免疫基因的表达情况,结果显示低浓度壳寡糖会激活炎症因子IL-6和IL-1β的表达,而高浓度壳寡糖会通过NF-κB途径抑制IL-6、IL-1β、TNF-α的表达。最后,通过哈维弧菌(Vibrio harveyi)攻毒实验检测了壳寡糖对珍珠龙胆石斑鱼的免疫保护作用,结果显示,壳寡糖能通过NF-κB细胞通路抑制炎症因子mRNA的表达,同时显著抑制了细胞凋亡相关基因的表达。综上所述,壳寡糖的添加可促进珍珠龙胆石斑鱼的生长,提高其非特异性免疫,具有一定的免疫保护作用。通过本研究,为壳寡糖在水产养殖中的应用提供理论参考,同时为珍珠龙胆石斑鱼的疫病防控提供依据。

      Oligochitosan (COS), are oligomers of glucosamine and N-acetyl glucosamine linked by β-1-4 glycosidic bonds, which exhibit biological activities such as anti-inflammation, antioxidation, growth promotion and so on. At present, Oligochitosan has been widely used in human or poultry health, but its application in aquatic products is still very lacking. In this paper, the immunomodulatory effect of Oligochitosan on peral gentian groupers (Epinephelus lanceolatu (× Epinephelus fuscoguttatus) was comprehensively analyzed by adding different doses of Oligochitosan (0, 200 mg/kg, 400 mg/kg and 800 mg/kg) to the basic experimental feed for four weeks, and the immunomodulatory effect of Oligochitosan on peral gentian groupers was analyzed comprehensively. In this study, the effects of Oligochitosan on the activities of immune and antibacterial related enzymes in peral gentian groupers were tested. Oligochitosan can significantly increase the activities of glutathione peroxidase, lysozyme, catalase, alkaline phosphatase, glutamic oxaloacetic transaminase and glutamic pyruvic transaminase in pearl gentian groupers. What’s more, the addition of Oligochitosan significantly increased the proportion of head kidney of pearl gentian groupers. The results of H E staining showed that compared with the control group, the head kidney tissue of Oligochitosan group was more compact, especially in 800 mg/kg group. In order to further explore the immunomodulatory effect of Oligochitosan on pearl gentian groupers, real-time quantitative PCR was used to detect the expression of immune genes in the head kidney of pearl gentian groupers. The results showed that low concentration of Oligochitosan could activate the expression of inflammatory cytokines IL-6 and IL-1 β, while high concentration of Oligochitosan could inhibit the expression of IL-6, IL-1 β and TNF- α through NF- κ B signaling pathway. Finally, the immunoprotective effect of Oligochitosan on pearl gentian groupers was tested by Vibrio harveyi challenge experiment. The results showed that Oligochitosan could inhibit the expression of inflammatory factor mRNA through NF- κ B signaling pathway, significantly inhibit the expression of apoptosis-related genes, and significantly improve the structural integrity of groupers head kidney tissue. To sum up, the addition of Oligochitosan can promote the growth of pearl gentian groupers, improve its non-specific immunity, and has a certain immune protective effect. This study provides a theoretical reference for the application of Oligochitosan in aquaculture and provides a basis for disease prevention of pearl gentian groupers.
